ABSTRACT

Objective: The study's goal has been to analyze if environmental or occupational exposure to pesticides can produce changes in pregnant women living in a countryside municipality. Methods: The participants of this study were twenty-three pregnant women, who both answered a questionnaire and donated biological material in order to perform Micronucleus (MN) Tests in lymphocytes, oral epithelial cells, and also for measuring the enzyme activity of erythrocyte acetylcholinesterase. Results: Considering the total analyzed samples, the following was found: an average of 8 ± 2.92 MN/1000 oral epithelial cells from urban participants; an average of 6.82 ± 3.43 MN/1000 oral epithelial cells from rural participants; and 100% of the microscope slides contained cells with two MN, which shows high intensity lesions to the DNA. There was found a high frequency of spontaneous abortions (34.8%), greater than in Brazil. Conclusion: The exposure of pregnant women living in a countryside municipality to pesticides may increase the rate of spontaneous abortions, as well as the chances of mutagenic effects


Objetivo: Analisar se a exposição ambiental ou ocupacional aos agrotóxicos causa alterações em gestantes residentes em um município rural. Métodos: Compuseram a amostra 23 gestantes, que responderam a um questionário e doaram amostras biológicas para a realização dos testes de micronúcleos (MN) em linfócitos, em células do epitélio oral, e para a dosagem da atividade da enzima acetilcolinesterase eritrocitária. Resultados: Obteve-se uma média de 8 ± 2,92 MN/1000 células do epitélio oral analisadas em amostras de participantes da zona urbana, 6,82 ± 3,43 MN/1000 de participantes da zona rural, e 100% das lâminas continham células com dois MN, o que demonstra lesões ao DNA de maior intensidade. Encontrou-se uma frequência elevada de casos de abortos espontâneos (34,8%), superior à encontrada no Brasil. Conclusão: A exposição de gestantes residentes em um município rural aos agrotóxicos eleva a taxa de abortos espontâneos, bem como as chances de ocorrência de efeitos mutagênicos

Resultados: Se obtuvieron una media de 8 ± 2,92 MN / 1000 células del epitelio oral analizadas en muestras de participantes de la zona urbana, 6,82 ± 3,43 MN / 1000 de participantes de la zona rural, y el 100% de las láminas contenían células con dos MN, lo que demuestra lesiones al ADN de mayor intensidad. Se encontró una frecuencia elevada de casos de abortos espontáneos (34,8%), superior a la encontrada en Brasil. Conclusión: La exposición de gestantes residentes en un municipio rural a los agrotóxicos eleva la tasa de abortos espontáneos, así como las posibilidades de ocurrencia de efectos mutagênicos

ABSTRACT

BACKGROUND Only benznidazole (Bnz) (1) and nifurtimox (Nfx) (2) are licensed for the treatment of Chagas disease although their safety and efficacy profile are far from ideal. Farmanguinhos from Fiocruz has developed seven nitroimidazole compounds (4-10) analogs of megazol (3). OBJECTIVES To evaluate whether the genotoxic effect of 3 was abolished in the seven nitroimidazoles (4-10) analogs using the in vitro alkaline comet assay (CA) and the in vitro cytokinesis-block micronucleus assay (CBMN) in whole human blood cells (WHBC) and correlate this effect with their trypanocidal activity using bloodstream trypomastigote forms of Trypanosoma cruzi. METHODS The toxicity of 3-10 to WHBC in the in vitro CA was determined using the fluorescein diacetate/ethidium bromide assay. DNA damage in the in vitro CA was evaluated according to tail size in four classes (0-3) and methyl methane-sulfonate (MMS) was used as a positive control. The cytotoxicity of 3-10 to WHBC in the CBMN was measured using the cytokinesis-block proliferation index and the replication index. The number of the micronucleate cells in 2,000 binucleate cells by experimental group was determined. Mitomycin C and N-deacetyl-N-methylcolchicine were used as positive controls. FINDINGS Compound 3 showed a significant DNA strand break effect through the in vitro CA and highly significant clastogenic and/or aneugenic effect in the CBMN. Compounds 5, 6, 8, 9 and 10 showed negative results in the CBMN and positive results in the in vitro CA, while the inverse effect was observed for 4 and 7. MAIN CONCLUSIONS Compound 10 was the most promising to proceed with the development as a drug candidate in the treatment of Chagas disease showing absence of chromosomal cytogenetic damage and high activity against T. cruzi, about two times higher than 3 and the clinical drug 1.

Introducción: La enfermedad cerebrovascular constituye un importante problema de salud a nivel mundial. En la actualidad se desarrollan investigaciones científicas dedicadas al estudio de los efectos del campo magnético de frecuencia extremadamente baja para su tratamiento. No es suficientemente clara la información acerca de su inocuidad en las dosis estudiadas. Objetivo: Estudiar la seguridad de la aplicación del campo magnético de frecuencia extremadamente baja a nivel del sistema nervioso central a través de un estudio toxicológico a dosis aguda, repetida y ensayo de micronúcleos en médula ósea. Métodos: Se conformaron tres grupos experimentales con ratas Sprague Dawley Cenp:SPRD jóvenes y sanas para los experimentos de toxicidad y ratones CENP: NMRI para la evaluación mutagénica. Se utilizaron controles negativos no tratados. En el ensayo de micronúcleos se incorporó un grupo control positivo al que se administró Ciclofosfamida por vía intraperitoneal. Se aplicó un campo magnético no homogéneo con niveles de inducción magnética de 6,5 y 15 mT, tomando como referencia el valor máximo sobre la superficie de la bobina. Para la aplicación del campo magnético la bobina estimuladora se colocó sobre la cabeza asegurando la exposición completa del encéfalo. Resultados: En ninguno de los ensayos se detectaron signos de toxicidad. Se comprobó así mismo que no se indujeron efectos genotóxicos ni citotóxicos sobre las células somáticas. Conclusiones: El tratamiento con campo magnético de frecuencia extremadamente baja a nivel del sistema nervioso central en las condiciones experimentales y dosis estudiadas es seguro(AU)


Introduction: Stroke is a major health problem all over the world. Nowadays are developed scientific researches devoted to the study of extremely low frequency magnetic field effects over this illness. The information about it safety is unclear yet. Objective: To study the safety of extremely low frequency magnetic field applied at central nervous system level wasby means ofa toxicological assay (Acute, repeated doses and micronucleus in bone marrow assay) Methods: Three experimental groups were made with Sprague Dawley Cenp: SPRD young and healthy rats for toxicity experiments and CENP: NMRI mice for mutagen evaluation. Untreated negative controls were used. In the micronucleus assay, an additional positive control group was included. This group received Cyclophosphamide by intraperitoneal administration. Was applied a non-homogenousmagnetic fieldof 6,5 and 15 mT, taken as reference the maximum value over the coil surface. The coil was positioned over the head, ensuring full exposure of brain to magnetic field. Results : In none of trials were detected any sign of toxicity. It was also found no genotoxic or cytotoxic effects induced on somatic cells. Conclusions : These results indicated the safety of treatmentwith extremely low frequency magnetic field at central nervous system level for experimental conditions and doses studied(AU)

ABSTRACT Tricyclazole is currently one of the fungicides recommended for the treatment of diseases in irrigated rice. However, there is relatively little information on its cytotoxic and genotoxic potential. The objective of this study was to evaluate the cytotoxicity and genotoxicity of rice crop water after apllication of the tricyclazole fungicide through the Allium cepa L. test. The rice crop water samplings were collected before and 1, 15 and 30 days after application of the fungicide in rice plant shoots. The Allium cepa roots were placed in contact with the rice crop water to check for possible chromosomal abnormalities and mitotic index of the bioindicators meristematic cells. The data obtained by the Allium cepa test indicates that the application of the tricyclazole fungicide leads to an increase in the genotoxic activity in the rice crop water, through the appearance of chromosomal abnormalities, without, however, causing significant effects on the mitotic index. The major chromosomal alterations observed were anaphasic and telophasic bridges and laggard chromosomes.

El Río Matanza-Riachuelo y sus afluentes atraviesan zonas con diferente grado de contaminación generada por las actividades agrícola-ganaderas, urbana e industrial. Los contaminantes que llegan al agua y son depositados en los sedimentos pueden ser liberados nuevamente al agua generando efectos tóxicos y/o genotóxicos sobre los organismos acuáticos. El objetivo de este trabajo fue analizar la genotoxicidad de muestras de sedimentos de la cuenca Matanza-Riachuelo obtenidas de zonas con diferentes usos del suelo. Se seleccionaron cuatro sitios de muestreo. Se utilizaron 2 métodos de extracción de contaminantes (agitación y sonicación), 2 solventes orgánicos (metanol y diclorometano) y 2 solventes inorgánicos (agua y solución ácida), obteniéndose un total de 5 extractos para cada muestra. Se realizaron mediciones de metales pesados e hidrocarburos aromáticos policíclicos (HAPs) mediante espectrofotometría de absorción atómica y CG/MS, respectivamente. La genotoxicidad se evaluó mediante el test de Ames con 2 cepas de Salmonella typhimurium (TA98 y TA100), con y sin fracción microsomal S9, y el test de Allium cepa. De los cuatro sitios estudiados, los sedimentos del Riachuelo mostraron mayores concentraciones de metales pesados y HAPs. Para el test de Ames, sólo los extractos obtenidos en diclorometano resultaron genotóxicos para la TA100 +S9 mix. Tanto los extractos inorgánicos como los orgánicos fueron citotóxicos y genotóxicos para A. cepa. Se observó una correlación negativa entre algunos compuestos HAPs y la frecuencia de micronúcleos, indicando la presencia de efectos antagónicos con otros compuestos genotóxicos. Los extractos con mayor efecto tóxico y genotóxico fueron los obtenidos con diclorometano y solución ácida. Este estudio mostró que los contaminantes orgánicos e inorgánicos extraídos de muestras de sedimento de la Cuenca Matanza-Riachuelo, con diferente grado de impacto, presentan un potencial riesgo tóxico y genotóxico para el ecosistema acuático.


The Matanza-Riachuelo River and its tributaries traverse areas with different degrees of contamination due to farming, urban and industrial activities. The pollutants entering the water are deposited in sediments, and can be released back into the water producing toxic and/or genotoxic effects on aquatic organisms. The aim of this study was to analyze the genotoxicity of sediment samples from the Matanza-Riachuelo Basin with different land uses. Four sampling sites according to the characteristics of land use were selected. Two methods of extraction (stirring and sonication), two organic solvents (methanol and dichloromethane) and two inorganic solvents (water and acid solution) were used, yielding a total of 5 extracts for each sample. Measurements of heavy metals and polycyclic aromatic hydrocarbons (PAHs) by atomic absorption spectrophotometry and GC/MS, respectively were performed. Genotoxicity was assessed using the Ames test with 2 strains of Salmonella typhimurium (TA98 and TA100) with and without S9 microsomal fraction, and the Allium cepa test. Taking into account the four sites, sediments from Riachuelo showed higher concentrations of heavy metals and PAHs. Only the dichloromethane extracts were genotoxic to the Ames test using the TA100 strain +S9 the mix. Both organic and inorganic extracts were cytotoxic and genotoxic to A. cepa. A negative correlation between some PAHs compounds and micronucleus frequency were observed, indicating the presence of antagonistic effects with other genotoxic compounds in samples. The extracts with high toxic and genotoxic effects were obtained with dichloromethane and acid solution. This study showed that organic and inorganic contaminants extracted from sediment samples from the Matanza-Riachuelo Basin, with varying degrees of impact, have potential toxic and genotoxic risk to the aquatic ecosystem.

O aumento na poluição ambiental é na atualidade uma das grandes preocupações em nível mundial. Especificamente, a poluição atmosférica por material particulado tem demostrado ser um fator determinante no desenvolvimento de doenças cardiopulmonares e câncer de pulmão nas populações expostas. O material particulado é constituído por uma mistura complexa de compostos orgânicos e inorgânicos, muitos dos quais possuem potencial mutagênico e genotóxico. As características destes compostos variam em função da suas propriedades físicas, químicas e em razão das condições meteorológicas prevalecentes. A maior parte dos estudos tem se focado em avaliar o potencial genotóxico da fração orgânica do material particulado e poucos estudos têm explorado a fração solúvel em água e a contribuição diferencial das diversas espécies químicas presentes nesta fração para o dano genotóxico. O objetivo deste trabalho foi avaliar os efeitos mutagênicos e genotóxicos in vitro da fração orgânica e da fração solúvel em água de material particulado (MP10) coletado em três locais diferentes do estado de São Paulo e estabelecer a relação entre a composição química e o efeito biológico observado. Para isto, realizou­-se a extração orgânica e solúvel em água de 12 amostras de MP10. A mutagenicidade e genotoxicidade foram avaliadas usando o ensaio de Salmonella/microssoma e o teste de micronúcleos (MN) em células A549 e 16 hidrocarbonetos Policíclicos Aromáticos (HPAs) e 15 metais hidrossolúveis presentes nas amostras foram determinados quimicamente. Adicionalmente, foi determinada a metodologia de extração da fracção solúvel em água e se avaliou a estabilidade química e biológica desta fração. Os resultados indicam que a extração assistida por micro-ondas é um método eficiente para a extração da fração solúvel em agua do MP e que um tempo superior a 60 dias de armazenamento e congelamento deste tipo de extrato tem um efeito significativo sobre os resultados analíticos e a resposta biológica. Foi demonstrado ainda que as duas frações de MP estudadas são responsáveis pela indução do dano ao DNA e que não existe uma relação direta entre a concentração de MP e o efeito genotóxico observado, confirmando a importância do uso de bioensaios na avaliação da genotoxicidade de misturas complexas como o MP. Os HPAs prevalecentes nas amostras de PM10 foram fluorantene e benzo(ghi)perileno. Nos extratos solúveis em água, as maiores concentrações de metais foram determinadas para zinco, ferro e cobre. Confirmou-se que a indução de MN e o ensaio de Salmonella/microssoma representam uma poderosa ferramenta na avaliação da polução atmosférica e que as análises químicas por si só não são suficientes para a proteção e predição dos efeitos biológicos em populações expostas


The increase of environmental pollution is today one a major concern worldwide. Specifically, air pollution by particulate matter has been shown to be a determining factor in the development of cardiopulmonary disease and lung cancer in exposed populations. The particulate material consists of a complex mixture of organic and inorganic compounds, many of which have mutagenic and genotoxic activity. The characteristics of these compounds vary according to their physical and chemical properties and also to the prevailing weather conditions. Most studies have focused on evaluating the genotoxic potential of the organic fraction of particulate material, but few studies have explored the water­-soluble fraction, and the differential contributions of different chemical species present in this fraction to genotoxic damage. The aim of this study was to evaluate the in vitro mutagenic and genotoxic effects of organic and water-soluble fractions of 12 samples of particulate matter (PM10) collected at three different sites in the state of São Paulo and establish the relationship between the chemical composition and the biological effect observed. The mutagenicity and genotoxicity were evaluated using the Salmonella/microssome test and the micronucleus assay (MN) in A549 cells and 16 Polycyclic Aromatic Hydrocarbons (PAHs) and 15 water-soluble metals present in the samples were chemically determined. Additionally, the extraction method of water- soluble fraction was determined and the chemical and biological stability of this fraction evaluated. The results indicate that the microwave­-assisted extraction is an efficient method for the extraction of the water-soluble compounds of PM and that the freezing and storage of the extract over 60 days has a significant effect on the mutagenic and analytical results of PM samples. It was demonstrated that the two PM fractions studied are responsible for the induction of DNA damage and that there is no direct relationship between the MP concentration and the genotoxic effect observed, confirming the importance of using bioassays in the genotoxicity evaluation of complex mixtures as PM. The PAHs prevailing in our samples were fluoranthene and benzo(ghi)perylene. In the water-soluble extracts, highest concentrations of the elements studied were found for zinc, iron, and copper in the three places of sampling. We confirmed that MN induction and Salmonella/microsome assay represents a powerful tool to evaluate the atmospheric air pollution and that the total concentration of PM and the chemical analyses alone would not be sufficient for the prognosis of biological effects in exposed populations

Human papillomavirus (HPV) has the highest mortality rate due to cervical cancer in Northeastern Argentina. The aim of this work was to detect and characterize HPV in samples from the Province of Corrientes, Argentina. HPV detection and typing was performed using PCR-RFLP on samples with different cervical lesions (n = 255). Seventeen viruses typified as HPV-58 were sequenced (E6 and E7 genes) and mutations were analyzed. HPV DNA was detected in 56.1 % of the cervical lesions (143/255). Twenty-two different HPV types were detected. The type most frequently found among the total number of samples and HPV-positive samples was HPV-16 (14.5 % and 25.9 %, respectively), followed by HPV-58 (8.2 %/14.7 %, respectively), which is also considered a high-risk viral type. Increased severity of the cytological status was associated with greater rates of HPV detection and, especially, with the detection of greater rates of high-risk types. In addition, the evolutionary dynamics of the alpha-9 species group and HPV-58 was studied. All HPV-58 viruses reported in this work belonged to lineage A, sublineage A2. The phylodynamic analysis indicated that diversification of main groups within lineage A might have accompanied or preceded human migrations across the globe. Given that the most prevalent viruses found belonged to high-risk HPV types, some concerns might arise about the extent of cross protection of the vaccines against the types not included in their design.


El virus del papiloma humano (Human papillomavirus [HPV]) tiene la mayor tasa de mortalidad por cáncer de cuello uterino en el noreste de Argentina. El objetivo de este trabajo fue detectar y caracterizar el HPV en muestras de la provincia de Corrientes, Argentina. La detección y la tipificación se realizó mediante PCR-RFLP en muestras con diferentes lesiones cervicales (n=255). Se secuenciaron 17 virus tipificados como HPV-58 (genes E6 y E7) y se analizaron sus mutaciones. Además, se estudió la dinámica evolutiva de los virus del grupo alfa-9 y, en particular, del HPV-58. Se detectó ADN viral en el 56,1% de las lesiones cervicales (143/255) y se detectaron 22 tipos del HPV. El tipo encontrado con mayor frecuencia entre el total de muestras y entre las HPV-positivas fue el HPV-16 (14,5%/25,9%, respectivamente), seguido por el HPV-58 (8,2%/14,7%, respectivamente), también considerado como de alto riesgo. El aumento de la gravedad de las lesiones se asoció a mayores tasas de detección del HPV y, en especial, con mayores tasas de detección de tipos de alto riesgo. Todos los HPV-58 encontrados pertenecieron al linaje A, sublinaje A2. El análisis filodinámico indicó que la diversificación de los grupos principales dentro del linaje A podría haber acompañado o precedido las migraciones humanas en todo el mundo. Dado que los virus más prevalentes pertenecieron a los tipos del HPV de alto riesgo, podrían surgir interrogantes sobre el alcance de la protección cruzada de las vacunas contra los tipos no incluidos en su diseño

ABSTRACT

O artigo analisa as estratégias de controle existentes no trabalho na mina de Morro Velho, Minas Gerais, e as mudanças resultantes da implementação da legislação trabalhista durante o governo Vargas. Discute as doenças causadas pelo trabalho na mina, silicose e arsenicismo, através de depoimentos de ex-mineiros e do livro de um autor anônimo que aborda as doenças e as relações de poder entre patrões e empregados, apontando os limites da legislação e das lutas operárias. O livro traz um depoimento contundente de como a empresa proprietária, inglesa, burlou, durante muito tempo, leis como a da taxa de insalubridade. Direito que outras mineradoras, não só de propriedade inglesa, costumavam e até hoje costumam desrespeitar pelo mundo.


This article analyzes the control strategies in place at Morro Velho mine in the Brazilian state of Minas Gerais, and the changes after the implementation of labor legislation during the Vargas administration. The diseases common amongst mine workers, silicosis and arsenicosis, are investigated through statements given by former miners and a book by an anonymous author that discusses the diseases and the power relations between employers and employees, identifying the limitations of the legislation and the workers’ struggles. The book presents a striking story of how for many years the British company side-stepped laws such as the insalubrity premium, a right which other mining companies, not only of British ownership, flouted and still flout in different parts of the world.

ABSTRACT

O bioensaio da mutação do pelo estaminal de Tradescantia clone 4430 (Trad-SHM) foi utilizado para avaliar a genotoxicidade de um herbicida composto por triazinas (atrazina e simazina) após exposição in situ. Trinta vasos da planta foram expostos durante a aplicação do herbicida (grupo teste) mantendo-se um grupo controle em casa de vegetação. A genotoxicidade foi expressa em termos de eventos mutantes pink (EMP) e a análise dos dados foi realizada por meio do teste t de Student em oito dias de avaliação (C8D = controle 8 dias; T8D = teste 8 dias) e no dia de pico (CPD = controle dia de pico; TPD = teste dia de pico). A exposição ao herbicida causou um número significativamente maior de EMP no grupo teste (T8D = 2,27; TPD = 4,69) do que no controle (C8D = 0,71; CPD = 0,62), demonstrando a existência de risco genotóxico associado ao uso das triazinas, sendo o bioensaio Trad-SHM uma eficiente ferramenta para avaliar o poten­cial genotóxico destes contaminantes ambientais causadores de efeitos adversos à saúde humana.


Tradescantia 4430 clone stamen hair mutation (Trad-SHM) bioassay was used to evaluate the genotoxicity of a herbicide composed of triazines (atrazine and simazine) after in situ exposure. Thirty plant pots were exposed during the herbicide application (test group) and a control group was maintained in a greenhouse (control group). Genotoxicity was expressed in terms of pink mutation events (EMPs) and the data analysis was performed by Student's t test comparing the control and contaminated group after eight-days (C8D = 8-day control; T8D = 8-day test) and peak day (CPD = peak day control; TPD = peak day test). Exposure to the herbicide caused a significantly higher number of EMPs in the test group (T8D = 2.27; TPD = 4.69) than in the control group (C8D = 0.71; CPD = 0.62). This demonstrates that the Trad-SHM bioassay is sensitive and efficient and can be used as tool to assess the genotoxic potential of environmental contaminants like triazines associated with adverse effects on human health.

O extrato aquoso de erva-mate, obtido a partir de folhas secas de Ilex paraguariensis, é uma bebida amplamente consumida na América do Sul. Inicialmente, nosso objetivo foi caracterizar os compostos presentes nas amostras de erva-mate disponíveis no mercado brasileiro (CH: chimarrão; T: chá mate torrado; G: chá mate torrado, comercialmente acondicionado em garrafas ou C: em copos; TS: chá mate torrado solúvel A mutagenicidade, citotoxicidade e antimutagenicidade de todas as amostras também foram avaliadas atavés do Teste de Ames na presença e na ausência de ativação metabólica. Em seguida, analisamos a amostra TS (2,5, 5,0 e 10 mg/mL) quanto a sua atividade antioxidante e antigenotóxica. Além disso, avaliamos também os efeitos da amostra TS sobre a sinalização da leptina e da insulina no hipotálamo e o estresse oxidativo hepático de ratos adultos obesos programados pela superalimentação neonatal (S). Para induzir S, o tamanho da ninhada foi reduzido a três filhotes por lactante e as ninhadas com número padrão de filhotes (dez/lactante) foram utilizadas como controle. Aos 150 dias de vida, as proles S foram subdivididas em: TS - tratados com extrato aquoso de erva-mate (1g/kg de peso corporal/dia, por gavagem) e S - recebendo água por gavagem durante 30 dias. A prole controle (C) também recebeu água nas mesmas condições do grupo S. Em nossos resultados, verificamos a presença de ácido clorogênico, cafeína e teobromina em todas as amostras analisadas. O conteúdo de compostos fenólicos nas infusões estudadas foram CH: 5,14±0,23; T: 4,33±0,01; G: 0,93±0,25; C: 0,80±0,3 e TS: 8,35±0,5 mg/ml. Não observamos efeito mutagênico ou citotóxico nas amostras analisadas. Um efeito antimutagênico significativo foi observado para a cepa TA97 (pré-, co- e pós-tratamento), na presença de ativação metabólica, em todas as amostras testadas. A amostra TS também apresentou um efeito antimutagênico significativo para a TA102 (pré-, co-e e pós-tratamento)...


Yerba mate extract, made from dried leaves of Ilex paraguariensis, is a tea-like beverage consumed in South America. Here, we aim, firstly, to characterize the compounds present in yerba mate beverages available in the Brazilian market (chimarrao –CH; mate tea –T; mate tea commercially packed in bottles –B and cups ready-to-drink –C; and roasted yerba mate soluble tea –TS). We also evaluated its mutagenic, cytotoxic and antimutagenic properties through Ames test in the presence and absence of metabolic activation. Then, we analyzed exclusively TS sample (2.5, 5.0 and 10 mg/mL) as its antioxidant activity, on plasmid DNA cleavages and upon epithelial esophagus primary culture of Wistar rats treated with a genotoxic agent. Futhermore, we evaluated the effects of mate tea (TS) upon the hypothalamic changes of leptin and insulin signaling and hepatic oxidative stress in postnatal early overfeeding (S) rats. To induce S, litter size was reduced to three pups per dam and normal litters (ten pups/dam) were used as control. In postnatal day (PN) 150, S offspring were subdivided into: S and TS groups treated, respectively, with water or mate tea (1g/kg BW/day, by gavage) during 30 days. C offspring received water. Our results showed the presence of chlorogenic acid, caffeine and theobromine in all analyzed samples. The contents of phenolic compounds in the studied infusions were CH:5,14±0,23; T:4,33±0,01; B:0,93±0,25; C:0,80±0,3 and TS:8,35±0,5 mg/mL. No mutagenic or cytotoxic effect was observed for all analyzed samples. A significant antimutagenic effect was observed for S.typhimurium TA97 (CoT, PreT and PosT), in the presence of metabolic activation, for all tested samples. TS sample also exhibited a significant antimutagenic effect for S.typhimurium TA102 (CoT, PreT and PosT) in the presence of metabolic activation. We did not observed an antimutagenic effect using TA98, TA100 and TA102 for CH, T, C and B samples; and no effect for TA98 and TA100 for TS sample...

BACKGROUND: Pesticides are used in agriculture to protect crops, but they pose a potential risk to farmers and environment. The aim of the present study is to investigate the relation between the occupational exposure to various pesticides and the presence of DNA damage. MATERIALS AND METHODS: Blood samples of 210 exposed workers (after a day of intense spraying) and 50 control subjects belonging to various districts of Punjab (India) were evaluated using Comet assay. Sixty workers who showed DNA damage were selected for follow up at 5-6 months after the first sampling during a low or null spraying period. RESULTS: Significant differences were found in DNA damage between freshly exposed workers and controls and freshly exposed and followed up cases. There was significant increase in the comet parameters viz. mean comet tail length and frequency of cells showing migration in exposed workers as compared to controls (72.22 ± 20.76 vs. 46.92 ± 8.17, P<0.001; 31.79 vs. 5.77, P<0.001). In the second samples, followed up cases showed significant decrease in frequency of damaged cells as compared to freshly exposed workers of first sampling (P<0.05). The confounding factors such as variable duration of pesticide exposure, age, smoking, drinking and dietary habits etc which were expected to modulate the damage, were instead found to have no significant effect on DNA fragmentation. CONCLUSION: The evidence of a genetic hazard related to exposure resulting from the intensive use of pesticides stresses the need for educational programs for agricultural workers to reduce the use of chemicals in agriculture.

ABSTRACT

Background: Approximately 15 percent of misoprostol-induced-abortions may not be successful, leading to in utero exposure to the drug and to the induction of a series of defects including central nervous system, limb and visceral defects. A commonproposal is that the drug causes disruption of the fetal vasculature leading to embryonic or fetal hypoxia. Aim: To evaluate the teratogenicity of misoprostol using the rat post-implantation embryo culture. Material and Methods: Rat embryos were collected at the beginning of organogenesis and cultured in rat serum containing misoprostol at concentrations of 200, 2,000 or 20,000 pg/ml. Functionality, morphology and morphometry parameters were evaluated. Results: Misoprostol induced a dose-dependent embryotoxic effect causing a decrease in embryo viability and function (poor vascular development and survival) and morphometry (alterations in branchial arches, heart and cephalic portions of the neural tube, among others). Conclusions: All the manifestations observed are indicative of the ability of misoprostol to directly induce developmental retardation and alterations.

BACKGROUND: For the past few decades, it has been widely known in developed countries that tobacco is dangerous, but it is still insufficiently realized how big these dangers really are. AIMS: To determine and evaluate micronuclei (MN) frequencies of young smokers and nonsmokers in three different tissues (peripheric blood lymphoctes, buccal mucosa, and exfoliative urothelial cells) at the same time. MATERIALS AND METHODS: MN assay was performed on buccal mucosa, urothelial cells, and peripheric blood lymphocyte samples obtained from 15 healthy male smokers (>5 pack-years) and 15 healthy male nonsmoker controls who had not been exposed to any known genotoxic agent. STATISTICAL ANALYSIS USED: The statistical differences between smoker and nonsmoker groups were calculated by using student t test. The differences between smoker-group tissues were compared by ANOVA. RESULTS: It was found that MN frequency (mean value ± standard deviation) in oral mucosa cells from smokers and controls were 1.20 ± 0.22% and 0.26 ± 0.10%; in urothelial exfoliative cells, 1.29 ± 0.28% and 0.12 ± 0.08%; in peripheric blood lymphocytes, 1.53 ± 0.23% and 0.38 ± 0.12%, respectively. The mean MN frequencies in buccal mucosa, urothelial exfoliative cells, and peripheric blood lymphocytes were significantly higher in smokers than in those of controls (P<0.05). All tissues were affected from smoking, but the most destructive effect was seen in urothelial cells of smokers (P<0.05). CONCLUSIONS: Our data showed that cigarette smoke is a DNA damage causitive agent on exfoliative buccal mucosa and urothelial cells and peripheric blood lymphocytes of young smokers, but it has most destructive effect on urothelial cells.

Plathymenia reticulata Benth has an anti-inflammatory effect and is capable of neutralizing the neuromuscular blockade induced by Bothrops jararacussu or Crotalus durissus terrificus venoms, probably by precipitating venom proteins (an effect caused by plant tannins). The present study aimed to evaluate the mutagenic activity of P. reticulata by using the Salmonella mutagenicity assay (Ames test) and the micronucleus test in CHO-K1 cells. P. reticulata extract concentrations of 2.84, 5.68, 11.37, and 19.90 mg/plate were assayed by the Ames test using TA97a, TA98, TA100 and TA102 bacterial strains, with (+S9) and without (-S9) metabolic activation. Concentrations of 5, 1.6 and 0.5 ìg/mL of P. reticulata extract were used for the micronucleus test. P. reticulata extract was mutagenic to TA98 (-S9) and showed signs of mutagenic activity in TA97a and TA102 (both -S9) strains. Micronucleus test CBPI values showed that the endogenous metabolic system increased the number of viable cells when compared to the non-activated samples and the micronucleus frequency increased when the cells were treated in the absence of S9. We concluded that P. reticulata extract may present direct mutagenic properties.

O cloreto estanoso (SnCI2) e a radiação ultravioleta A (UVA) são agentes que lesam diversas estruturas celulares, inclusive o DNA, principalmente pela geração de espécies reativas de oxigênio. O objetivo deste trabalho foi estudar a mutagênese e o reparo das lesões produzidas pela combinação do UVA, na condição de pré-iluminação, com o SnCI2. Avaliou-se a ação de enzimas do reparo por excisão de bases (BER), em Escherichia coli (E. coli), por eletroforese em gel alcalino de agarose e sobrevivência bacteriana. Também se estudou a indução do sistema SoxRS pelo cromoteste, e a mutagênese pelo teste de Ames. De acordo com os resultados: i)o UVA induziu quebras no DNA das cepas testadas e os mutantes fpg-nfo e fpg apresentaram maior retardo no reparo das lesões; ii) o SnCI2 induziu mais quebras que o UVA e os mutantes nfo e fpg mostraram maior dificuldade em reparar as lesões; iii) o UVA+SnCI2 provocou mais quebras que o SnCI2 e os mutantes nfo e fpg também apresentaram maior lentidão no reparo das lesões; iv) o UVA não inativou as cepas testadas; v) as cepas nfo e fpg foram as mais sensíveis ao SnCI2; vi) o UVA+SnCI2 provocou maior letalidade em todas as cepas testadas, em relação ao SnCI2, e os mutantes nfo e fpg também foram os mais sensíveis ao tratamento com ambos os agentes; vii) a transformação dos mutantes nfo com o plasmídio pBW21 (nfo+) e dos mutantes fpg com o plasmídio pFPG (fpg+) aumentou a sobrevivência das cepas aos tratamentos com SnCI2 e UVA+SnCI2; viii) o SnCI2 induziu o sistema SoxRS; ix) o SnCI2, UVA e UVA+SnCI2 não induziram mutagênese; x) o reparo das lesões parece ser preferencialmente realizado pelas proteínas Fpg e Nfo.


Stannous chloride (SnCI2) and ultraviolet radiation A (UVA) are able to induce lesions in different cellular structures, including DNA, manly through ROS generation. The aim of this work was to study the mutagenesis and repair of lesions induced by the association of UVA (pre treatment) with SnCI2. It was evaluated the action of base excision repair (BER) enzymes in Escherichia coli (E. coli) by alkaline gel electrophoresis and bacterial survival. It was also evaluated the SoxRS system induction by chromotest and mutagenesis through the Ames test. According to the results: i) UVA induced DNA strand breaks in all strains and fpg-nfo and fpg mutants showed greater delay in the repair of lesions; ii) SnCI2 induced more breaks than UVA and nfo and fpg mutants showed more difficult to repair the damage; iii) UVA + SnCI2 caused more breaks than the SnCI2 and nfo and fpg mutants also showed a slowest repair of injuries; iv) UVA did not inactivate any bacterial strains tested; v) nfo and fpg strains were more sensitive to SnCI2; vi) UVA + SnCI2 caused higher mortality in all strains tested, when compared to SnCI2, and, again, nfo and fpg mutants were the most sensitives to the treatment with both agents; vii) the transformation of nfo mutant with the plasmid pBW21 (nfo+) and fpg mutants with plasmid pFPG (fpg+) increased the survival of the strains to SnCI2 and UVA + SnCI2 treatments; viii) SnCI2 was able to induce SoxRS system; ix) SnCI2, UVA + SnCI2 and UVA did not induce mutagenesis; x) damage repair seems to be preferentially performed by Fpg and Nfo proteins.

ABSTRACT

The genotoxic effect of the Austroplenckia populnea chloroform fraction from barkwood extract was tested in vivo on peripheral blood cells of Swiss mice with the comet assay (SCGE), and the clastogenic effect was investigated on peripheral blood cells of Swiss mice and bone marrow cells of Wistar rats, with the micronucleus and chromosome aberrations tests. The animals were treated by gavage with 3 concentrations of the extract: 300, 600 and 900 mg.kg-1. Peripheral blood cells of Swiss mice were collected 4 and 24 hours after the treatment to the SCGE assay and 48 and 72 hours to the micronucleus test. Bone marrow cells of Wistar rats were collected 24 hours after the treatment to the micronucleus and chromosome aberration tests. The results showed that the A. populnea chloroform fraction induced an increase in the average number of DNA damage in peripheral blood cells at the three concentrations tested, but this increase was not statistically significant. In the micronucleus and chromosome aberrations test, no significant increase was observed in the mean number of micronucleated polychromatic erythrocytes (MNPCE) of Swiss mice or MNPCE or chromosome aberrations for the rat bone marrow cells, for any of the tested doses. Our findings enable us to conclude that by the comet assay, A. populnea chloroform fraction from barkwood extract showed no genotoxic effects, and by the micronucleus and chromosome aberration tests, the extract fraction showed no clastogenic/aneugenic effects on the rodent cells tested.


O possível efeito genotóxico da fração clorofórmica de cascas de caule de Austroplenckia populnea foi testado in vivo em células do sangue periférico de camundongos Suíços pelo ensaio cometa, e o seu possível efeito clastogênico foi investigado em células de sangue periférico de camundongos Suíços e células da medula óssea de ratos Wistar, respectivamente pelos testes do micronúcleo e de aberrações cromossômicas. Os animais foram tratados por via oral com três concentrações do extrato: 300, 600 e 900 mg.kg-1 de peso corpóreo. Células do sangue periférico dos camundongos foram coletadas 4 e 24 horas após o tratamento para a realização do ensaio cometa, e 48 e 72 horas para o teste do micronúcleo. Células da medula óssea dos ratos Wistar foram coletadas 24 horas após o tratamento para os testes do micronúcleo e de aberrações cromossômicas. Os resultados mostraram que a fração clorofórmica do extrato de A. populnea induziu um pequeno aumento no número médio de danos ao DNA nas células sanguíneas nas três concentrações testadas, mas tal aumento não foi estatisticamente significativo. Nos testes do micronúcleo e de aberrações cromossômicas não houve um aumento significativo no número médio de eritrócitos policromáticos micronucleados (EPM) dos camundongos, bem como nos EPM e aberrações cromossômicas nas células da medula óssea dos ratos, em nenhuma das doses testadas. Nossos resultados nos permitem concluir que, pelo ensaio cometa, a fração clorofórmica de cascas do caule de A. populnea não apresentou efeito genotóxico, e, pelos testes do micronúcleo e de aberrações cromossômicas, o extrato não mostrou efeitos clastogênicos e/ou aneugênicos nas células analisadas dos roedores.

O presente trabalho foi organizado em três capítulos. O objetivo do capítulo 1 foi avaliar a presença de atividade mutagênica em amostras de alguns poços do Estado de São Paulo, utilizando o ensaio Salmonella/microssoma. A atividade mutagênica foi avaliada utilizando três métodos diferentes: a) extração com resina XAD-4 em pH natural e ácido e b) extração líquido-líquido em pH básico, neutro e ácido, ambos seguidos do teste em microssuspensão (Kado) com as linhagens TA98 e TA100 de Salmonella thyphimurium na presença e ausência de mistura 89; c) análise de água não concentrada, apenas filtrada em membrana de 0,451..1m seguida do teste Salmonella/microssoma com pré- incubação com as linhagens TA98, TA100, YG1041 e YG1042, na presença e ausência de mistura S9. Foram avaliados nove locais diferentes em três campanhas. As amostras não concentradas não apresentaram atividade mutagênica, com as quatro linhagens testadas. Em relação aos métodos de extração, a melhor resposta foi obtida para o método XAD-4 seguido do teste em microssuspensão, e das 17 amostras avaliadas, quatro apresentaram atividade mutagênica. As respostas variaram nas diferentes campanhas para um mesmo poço, por isso outras análises nos mesmos locais devem ser realizadas, para uma melhor compreensão dos resultados obtidos. Os locais que apresentaram atividade mutagênica foram correspondentes às regiões mais vulneráveis. Os dados sugerem que o teste Salmonella/microssoma pode ser utilizado como um importante instrumento de triagem para monitorar a qualidade das águas subterrâneas

ABSTRACT

Adult lead poisoning commonly occurs from exposure to lead used in workplace. Lead exposure can cause a wide spectrum of adverse health effects. Its genotoxic effects are still debatable. To measure the adverse effects of lead exposure in printing house on some hematological parameters [red blood cells count, hemoglobin, and hematocrit levels] of workers, and to investigate its genotoxic effects on their leukocytes. The study was carried out on 40 workers exposed to lead in their work at Menoufiya University printing house, and ten control subjects selected from an area free from lead exposure and matched for age and smoking habit. An ex-post-facto study design was used. An interview questionnaire was used to assess biosocial characteristics, occupational history, and work related symptoms. Blood lead, red blood cells [RBCs], hemoglobin [Hb], and hematocrit levels [Hct] were evaluated. DNA fragmentation assay was used for detection of DNA fragmentation in leukocytes. Lead exposed workers has significantly higher mean blood lead than that controls [p < 0.001], 26.4 +/- 7.1 micro g/dL and 8.7 +/- 4.1 micro g/dL, respectively. Exposed workers has significantly lower mean RBCs [4.7 +/- 0.4], Hct [38.2 +/- 4.2] and Hb [13.9 +/- 0.7], compared to controls 5.2 +/- 0.5, 41.3 +/- 4.1, and 14.6 +/- 1.5, respectively. Statistically significantly higher frequency of symptoms of metallic taste, weight loss, fatigue, lack of memory, visual disturbance, and joint pain were reported by exposed workers, compared to controls. Statistically significant increased mean of optical density of DNA fragments at 200, 400 and 600 base pair of leukocytes of lead exposed workers were also shown, compared to controls. Statistically significant negative correlations were revealed between blood lead, duration of work and RBCs, Hct, and Hb. Statistically significant positive correlations were also revealed between blood lead, duration of work and total mean of optical density of DNA fragments. Occupational lead exposure, with increased blood lead level, is associated with decreased RBCs, Hct, and Hb and increased DNA fragmentation in leukocytes of printing workers. It is also associated with increased frequency of many related reported symptoms. Occupational health sendees are essential to eliminate the hazardous effect of lead exposure

The objective of this study was to document teratogenicity observed in chick embryos following administration of insecticide malathion in a dose of 2mg in 0.1 ml corn oil, and to suggest reasonable explanations for these anomalies. A total number of 300 eggs of Gallus domesticus species were used. After 48 hours of incubation eggs were divided into 5 groups, of 60 eggs each. The individual groups were subdivided into control [20], and treated [40] eggs. The control eggs were injected with 0.1 ml of corn oil, while the treated eggs were injected with 0.1 ml of corn oil in which 2 mg of malathion were dissolved. Eggs of both control and treated groups were examined at the 5[th], 7[th], 10[th], 14[th] and 18[th] days of incubation, for weight, mortality and morbidity, external malformations and body measurements. Embryos were prepared for skeletal examination with Alizarin red stain and Victoria blue stain. It's observed from the present study that lethality; external malformations and growth retardation, are characteristic features for malathion toxicity in chick embryo. It is observed that, malathion mortality is more frequent in higher age groups [14[th] and 18[th] days of incubation] while teratogenicity is more frequent in younger age groups [5[th] and 7[th] day of incubation]. Significant loss of weight in the treated groups is also observed. The characteristic external malformations were in the form of short lower peak, parrot beak, short neck, wry neck, micromelia of both fore limbs and hind limbs. In addition, tibiotarsal angulations and claw toes were also observed. Abnormal feather distribution, persistence of mesencephalic bulge, eye anomalies and visceral herniation could also be detected. It is concluded from this study that malathion injection is teratogenc in chick embryo when given in the 2[nd] day of incubation. The lethality detected in older age groups could be explained to be secondary to marked teratogenicity in vital organs such as heart [congestive heart failure] or neural tube defects. The toxicity of malathion on developing chick embryo could be explained by its anticholinesterase action or its suppressive effect on nicotinamid dinucleotide [NAD] levels. Also, its genotoxicity or mutagenicity could not be excluded
